Brand Name :
Gynazole 1, Femstat 3
Synonyms :
butoconazole
Class :
Antifungal, Vaginal anti-infective
Dosage Forms & Strengths
vaginal cream
2%
5g of the cream (1 applicatorful of cream) administered intravaginally as a single dose
Extend the dose for 6 days only in the second and third trimesters of pregnancy (Not recommended in the first trimester)
Safety and efficacy not established
Refer adult dosing
may decrease the therapeutic effect of antifungal agents
Mechanism of action
The mechanism of action of butoconazole is based on its ability to inhibit the synthesis of ergosterol, a critical component of fungal cell membranes. This results in a loss of membrane function and leads to the death of the fungal cells
Spectrum
The spectrum of activity of butoconazole is primarily against Candida species, including Candida albicans, which is the most common cause of vaginal yeast infections. It is also effective against some other species of Candida and other fungal pathogens. It is not active against bacteria or viruses
Frequency undefined:
vaginal irritation
vaginal swelling
abdominal pain
Contraindications
Caution
Pregnancy consideration: C
Lactation: Excretion of the drug in human breast milk is unknown
Pregnancy category:
Category A: well-controlled and Satisfactory studies show no risk to the fetus in the first or later trimester.
Category B: there was no evidence of risk to the fetus in animal studies, and there were not enough studies on pregnant women.
Category C: there was evidence of risk of adverse effects in animal reproduction studies, and no adequate evidence in human studies must take care of potential risks in pregnant women.
Category D: adequate data available with sufficient evidence of human fetal risk from various platforms, but despite the potential risk, and used only in emergency cases for potential benefits.
Category X: Drugs listed in this category outweigh the risks over benefits. Hence these categories of drugs need to be avoided by pregnant women.
Category N: There is no data available for the drug under this category
Pharmacology
It is an imidazole antifungal drug used to treat vaginal yeast infections caused by Candida species. It works by inhibiting the synthesis of ergosterol, a component of fungal cell membranes, which leads to a loss of membrane function and death of the fungal cells
Pharmacodynamics
The pharmacodynamics of butoconazole is based on its ability to inhibit the synthesis of ergosterol, a component of fungal cell membranes, by binding to the fungal cytochrome P450 14α-demethylase enzyme. This results in a loss of membrane function and death of the fungal cells
