Contraindications:Â
Caution:Â
Pregnancy consideration: There was evidence of risk of adverse effects in animal reproduction studies, and no adequate evidence in human studies must take care of potential risks in pregnant women. Â
Pregnancy category:Â CÂ
Lactation: The drug is not excreted in human breast milk Â
Pregnancy category:Â
Category A: well-controlled and Satisfactory studies show no risk to the fetus in the first or later trimester.Â
Category B: there was no evidence of risk to the fetus in animal studies, and there were not enough studies on pregnant women.Â
Category C: there was evidence of risk of adverse effects in animal reproduction studies, and no adequate evidence in human studies must take care of potential risks in pregnant women.   Â
Category D: adequate data with sufficient evidence of human fetal risk from various platforms, but despite the potential risk, and used only in emergency cases for potential benefits.   Â
Category X: Drugs listed in this category outweigh the risks over benefits. Hence these categories of drugs need to be avoided by pregnant women.   Â

Pharmacology:Â
oral mucoadhesive formulations contain polymers or bioadhesive agents that adhere to the mucosal surfaces in the oral cavity. This adhesion is facilitated by the hydration and swelling of mucoadhesive polymers upon contact with saliva. mucoadhesion extends the residence time of the formulation on the mucosal surfaces, promoting sustained contact and controlled drug release.Â
Pharmacokinetics:Â
AbsorptionÂ
mucoadhesive formulations can enhance drug absorption by increasing the residence time at the absorption site (e.g., oral mucosa). This can result in improved drug bioavailability. Drugs may be absorbed through the mucosal membrane, bypassing the gastrointestinal tract and potentially avoiding first-pass metabolism.Â
DistributionÂ
mucoadhesive formulations are designed to act locally at the site of administration. Distribution may be limited to the oral cavity or may lead to systemic distribution, depending on the drug properties and formulation.Â
MetabolismÂ
For drugs that undergo significant first-pass metabolism in the liver after oral administration, the mucoadhesive route might reduce exposure to hepatic metabolism, potentially altering the overall metabolic fate of the drug.Â
Elimination and ExcretionÂ
Depending on the drug’s properties, excretion may occur locally in the oral cavity through saliva or systemically through the kidneys or other organs.Â
Administration:Â
Oral administrationÂ
Mouthwashes or Rinses:Â
mucoadhesive polymers can be included in mouthwashes or rinses for local application to the oral mucosa.Â
The formulation may adhere to the mucosa, providing prolonged contact.Â
