Black Box Warning:
If pregnancy is detected, discontinue the dose as soon as possible. The drug affects the RAS, causing oligohydramnios. It may result in injury/death of the fetus.

Contraindications:
Contraindication in pregnancy: quinapril/hydrochlorothiazide is contraindicated during pregnancy, as it can cause harm to the developing fetus.
Contraindication in patients with a history of angioedema: quinapril/hydrochlorothiazide is contraindicated in patients with a history of angioedema, a condition characterized by swelling of the face, lips, tongue, and throat. It is because quinapril can increase the risk of angioedema, which can be life-threatening in some cases.

Pregnancy category:

Category C: there was evidence of risk of adverse effects in animal reproduction studies, and no adequate evidence in human studies must take care of potential risks in pregnant women.
Category D: adequate data available with sufficient evidence of human fetal risk from various platforms, but despite the potential risk, and used only in emergency cases for potential benefits.

Pharmacology:
quinapril works by inhibiting the action of the angiotensin-converting enzyme (ACE), an enzyme that converts angiotensin I to angiotensin II. Angiotensin II is a potent vasoconstrictor that causes blood vessels to narrow, which increases blood pressure. By blocking the action of ACE, quinapril reduces the levels of angiotensin II, leading to vasodilation (widening of blood vessels) and a decrease in blood pressure.
Pharmacodynamics:
quinapril is an angiotensin-converting enzyme (ACE) inhibitor. ACE is an enzyme that converts angiotensin I to angiotensin II, a potent vasoconstrictor that increases blood pressure by causing the blood vessels to narrow. quinapril inhibits the activity of ACE, which leads to a decrease in the levels of angiotensin II. This, in turn, causes vasodilation, or the widening of the blood vessels, leading to a decrease in blood pressure.
hydrochlorothiazide is a thiazide diuretic that works by inhibiting the reabsorption of sodium and chloride in the kidneys. It leads to an increase in urine output and a decrease in blood volume, which reduces the workload on the heart and blood vessels and decreases blood pressure.

Administration:
quinapril/hydrochlorothiazide is taken once daily, with or without food. The tablet should be swallowed whole with a full glass of water.
