Guacetisal is synthesized from aspirin and guaiacol by esterification reaction.
Pharmacodynamics:
It blocks the prostaglandin synthesis by inhibiting the activity of the enzyme cyclooxygenase. It is also known to suppress cough by acting on the medullary cough centre

Absorption
Following oral administration, the drug is absorbed rapidly in the g.i.tract, and the unchanged form is seen in the blood.
Metabolism
Undergoes hepatic metabolism to guajacholic ester of salicylic acid by esterases. This is, inturn, broken down into guajacol and salicylic acid by hepatic esterases.
Elimination and excretion
Guacetisal is excreted renally.
