Actions and Spectrum:Â
Action:Â
Analgesic action: Nifenazone aids in the relief of pain by lowering prostaglandin synthesis, which is involved in the transmission of pain signals; this action makes it effective in managing mild to moderate pain.Â
Anti-inflammatory action: Nifenazone decreases inflammation by inhibiting the formation of prostaglandins, which are mediators of the inflammatory response. This property makes it useful in conditions involving inflammation, such as arthritis.Â
Black Box Warning:Â Â
None
Contraindication/Caution:Â Â
Hypersensitivity: Individuals with known hypersensitivity or allergic reactions to nifenazone or any of its components should avoid its use.Â
Gastrointestinal Disorders: People with a history of gastrointestinal bleeding, ulcers, or perforation related to NSAID use should avoid nifenazone due to the potential for exacerbating these conditions.Â
Severe Heart Failure: Nifenazone can cause fluid retention and edema, potentially worsening heart failure symptoms. Therefore, it’s generally avoided in individuals with severe heart failure.Â
Severe Renal or Hepatic Impairment: Patients with severe kidney or liver problems should avoid nifenazone, as it may further impair these organs’ function.Â
Bleeding Disorders: Nifenazone, like other NSAIDs, can inhibit platelet function and may enhance bleeding more likely in people with bleeding disorders or those using anticoagulant drugs.Â
Pregnancy and Lactation: Nifenazone may harm the fetus or cause labor difficulties. Consequently, it is usually avoided when pregnant, especially in the third trimester. Additionally, it’s not recommended for use while breastfeeding due to potential adverse effects on the infant.Â
Pregnancy warnings:    Â
Pregnancy category: N/AÂ
Lactation: Excreted into human milk is unknownÂ
Pregnancy Categories:        Â
Category A: Studies that were well-controlled and met expectations revealed no risk to the fetus in either the first or second trimester.Â
Category B: There were a lack of studies on pregnant women and no evidence of risk to the fetus in animal experiments.  Â
Category C: there was evidence of risk of adverse effects in animal reproduction studies, and no adequate evidence in human studies must take care of potential risks in pregnant women.   Â
Category D: adequate data with sufficient evidence of human fetal risk from various platforms, but despite the potential risk, and used only in emergency cases for potential benefits.  Â
Category X: Drugs listed in this category outweigh the risks over benefits. Hence these categories of drugs need to be avoided by pregnant women.   Â
Category N: There is no data available for the drug under this category
Pharmacology: Nifenazone is a nonsteroidal anti-inflammatory drug with analgesic, anti-inflammatory, and antipyretic properties. It belongs to the class of pyrazolone derivatives.Â
Pharmacodynamics:Â Â
Anti-inflammatory Effects: Nifenazone exerts its anti-inflammatory effects by suppressing the production of prostaglandins, thereby decreasing the inflammatory reaction linked to conditions like arthritis, musculoskeletal disorders, and other inflammatory conditions.Â
Analgesic Properties: By inhibiting prostaglandin synthesis, nifenazone helps alleviate pain associated with various conditions, including mild to moderate pain, musculoskeletal pain, and postoperative pain.Â
Pharmacokinetics:Â
AbsorptionÂ
Nifenazone is well absorbed after oral administration. It is quickly absorbed from the digestive system, reaching peak plasma concentrations in one to two hours after consumption.Â
DistributionÂ
It has a moderate volume of distribution. Nifenazone is extensively bound to plasma proteins, mainly albumin. Â
MetabolismÂ
Metabolism of nifenazone primarily occurs in the liver. It undergoes extensive hepatic metabolism via various cytochrome P450 enzymes, primarily CYP1A2 and CYP2C9, to form several metabolites.Â
Excretion and EliminationÂ
The primary route of excretion for nifenazone and its metabolites is through the urine.
Administration: Â
The route of administration is oral.Â
The healthcare provider determines the frequency and dose based on the condition being treated.Â
