Contraindication / Caution:
Contraindications:
Pregnancy: Estetrol is contraindicated during pregnancy. It should not be used as an oral contraceptive or for any other purpose if the individual is already pregnant.
Breast Cancer: Estetrol is contraindicated in individuals with a history of breast cancer or current breast cancer. It may stimulate the growth of breast cancer cells.
Liver Disease: Individuals with severe liver disease or a history of liver tumors should avoid estetrol, as it may affect liver function.
Precautions:
Smoking: Smoking increases the risk of serious cardiovascular side effects associated with oral contraceptives. Estetrol users who smoke should be cautious and consider alternative contraception methods.
High Blood Pressure: Estetrol can affect blood pressure, and individuals with hypertension should be monitored carefully while using it.
Thromboembolic Disorders: Estetrol, like other hormonal contraceptives, may increase the risk of blood clots. It should be used with caution in individuals with a history of thromboembolic disorders.
Migraines: Individuals with a history of severe or recurrent migraines should use estetrol with caution.
Gallbladder Disease: There is an increased risk of gallbladder disease with the use of oral contraceptives, including those containing estetrol.
Diabetes: Individuals with diabetes should monitor their blood sugar levels carefully while using estetrol, as it can affect glucose tolerance.
Cholestatic Jaundice: Estetrol should be used with caution in individuals with a history of cholestatic jaundice associated with prior contraceptive use.
Depression: Mood changes and depression have been reported with hormonal contraceptives. Individuals with a history of depression should be cautious.

Lactation:Â Excreted into human milk is unknown
Pregnancy Categories:
Category A:Â Studies that were well-controlled and met expectations revealed no risk to the fetus in either the first or second trimester.
Category B: There were a lack of studies on pregnant women and no evidence of risk to the fetus in animal experiments.
Category C:Â there was evidence of risk of adverse effects in animal reproduction studies, and no adequate evidence in human studies must take care of potential risks in pregnant women.
Category D:Â adequate data with sufficient evidence of human fetal risk from various platforms, but despite the potential risk, and used only in emergency cases for potential benefits.
Category X:Â Drugs listed in this category outweigh the risks over benefits. Hence these categories of drugs need to be avoided by pregnant women.
Category N:Â There is no data available for the drug under this category

Why do we use estetrol?
estetrol is a natural estrogenic steroid hormone that is present in humans, alongside other estrogens like estrone, estradiol, and estriol. It is uniquely produced by the fetal liver and is detectable primarily during pregnancy. During pregnancy, it is found in relatively high levels in the fetus and lower levels in the mother’s circulation.
In the medical field, estetrol serves several purposes. It has gained approval as a new estrogenic component of combined oral contraceptives (COC). Additionally, it is undergoing clinical development for addressing menopausal symptoms and for potential use in managing breast and prostate cancer.
In the context of contraception, estetrol is used in combination with drospirenone, a well-known progestin. The synergy between these hormones suppresses ovulation, which is the primary mechanism by which they prevent pregnancy.
