Demeclocycline prevents cell division by blocking translation. It stops the amino acyl tRNA from attaching to the ribosome’s A site by binding (reversibly) to the 30S and 50S ribosomal subunits. 

Since many tetracycline antibiotics might result in diabetes insipidus, their use in SIADH depends on this side effect.

Lactation: Demeclocycline can be excreted in breast milk and may cause tooth discoloration in infants. Avoid use during breastfeeding.  

Pregnancy category: 

  • Category A: Satisfactory and well-controlled studies show no evidence of risk to the fetus in the first trimester or the later trimester. 
  • Category B: There is no evidence of risk to the fetus found in animal reproduction studies and there are not enough studies on pregnant women. 
  • Category C: Adverse effects on the fetus found with evidence in animal reproduction studies and no adequate evidence for an effect in humans, care must be taken for potential risks in pregnant women. 
  • Category D: There is adequate data available with sufficient evidence of human fetal risk from various platforms, but despite potential risks may be used only in emergency cases for potential benefits.

The lipophilic nature of demeclocycline allows it to passively diffuse through bacterial membrane porin channels or readily cross cell membranes. 

It is a bacteriostatic medication that inhibits the growth of microorganisms. It keeps effective blood levels longer than tetracycline because it is eliminated more gradually. 

Demeclocycline is not a direct bactericidal agent while it is a bacteriostatic drug. 

Administer 1 hour prior to or 2 hours after meals. 

Avoid taking it with antacids, multivitamins, calcium or magnesium supplements, iron supplements, or milk.

Demeclocycline prevents cell division by blocking translation. It stops the amino acyl tRNA from attaching to the ribosome’s A site by binding (reversibly) to the 30S and 50S ribosomal subunits. 

Since many tetracycline antibiotics might result in diabetes insipidus, their use in SIADH depends on this side effect.

Pregnancy considerations: demeclocycline can be assigned under pregnancy category D. Avoid during pregnancy. 

Lactation: Demeclocycline can be excreted in breast milk and may cause tooth discoloration in infants. Avoid use during breastfeeding.  

Pregnancy category: 

  • Category A: Satisfactory and well-controlled studies show no evidence of risk to the fetus in the first trimester or the later trimester. 
  • Category B: There is no evidence of risk to the fetus found in animal reproduction studies and there are not enough studies on pregnant women. 
  • Category C: Adverse effects on the fetus found with evidence in animal reproduction studies and no adequate evidence for an effect in humans, care must be taken for potential risks in pregnant women. 
  • Category D: There is adequate data available with sufficient evidence of human fetal risk from various platforms, but despite potential risks may be used only in emergency cases for potential benefits. 
  • Category X: Drugs listed in this category outweigh risks over benefits. These category drugs should be prohibited for pregnant women. 
  • Category N: There is no data available for the drug under this category. 

The lipophilic nature of demeclocycline allows it to passively diffuse through bacterial membrane porin channels or readily cross cell membranes. 

It is a bacteriostatic medication that inhibits the growth of microorganisms. It keeps effective blood levels longer than tetracycline because it is eliminated more gradually. 

Demeclocycline is not a direct bactericidal agent while it is a bacteriostatic drug. 

Administer 1 hour prior to or 2 hours after meals. 

Avoid taking it with antacids, multivitamins, calcium or magnesium supplements, iron supplements, or milk.

It is also indicated in management of urinary tract infections, gum disease, malaria.
